宋锋玲大连理工大学精细化工国家重点实验室教授,博士生导师,首批国家自然科学基金优秀青年项目获得者,教育部新世纪优秀人才计划入选者,大连市杰出青年科技人才培育计划入选者。主要从事有机荧光染料的结构性能与应用研究,并开展荧光染料及功能化荧光纳米粒子在生物医学领域的成像研究。研究成果获2007年全国百篇优秀博士学位论文提名奖; 2012年教育部自然科学一等奖;2012年辽宁省自然科学一等奖;2016年辽宁省自然科学学术成果论文类一等奖。

精细化工国家重点实验室彭孝军院士团队宋锋玲老师一直致力于有机荧光染料和荧光纳米粒子的光物理现象、及其在生物荧光成像等生物医学领域的创新应用研究。在最近的研究中,他们基于前期工作(J. Mater. Chem. B, 2017, 5, 5278-5283; ACS Appl. Mater. Interfaces, 2019, 11, 25750-25757; Chem. Commun., 2019, 55, 14522-14525),提出了基于分子工程和纳米材料改性的组合新策略,双管齐下,攻克了传统有机荧光染料在水性介质中的ACQ和荧光漂白的难题,实现了FSNPs在肿瘤细胞内长时间、高亮荧光成像和稳定的PDT过程。即通过分子工程策略对共价掺杂的近红外氨基菁染料进行分子结构优化设计、并结合纳米材料改性策略在FSNPs内部构建“纳米疏水笼”结构提供了一个相对非极性微环境,来同时攻克FSNPs发生在水性介质中的上述缺陷。研究人员通过改性的反相微乳液法制备了一系列掺杂不同结构菁染料和正辛基三乙氧基硅烷(OTES)改性的FSNPs。通过对其在水中荧光寿命、相对荧光强度和抗光漂白等光物理性能的系统分析,验证了增加染料分子中共价掺杂锚定位点和位阻基团数量、以及OTES构建的“纳米疏水笼”结构对强化FSNPs光物理性能的积极作用。即通过增加共价掺杂锚定位点和位阻基团数量使得染料分子在纳米粒子内部运动受限而减少非辐射跃迁能量衰变,同时又避免了因π−π堆积而导致的ACQ现象。于此同时,利用OTES分子内烷基链的疏水特性构建的“纳米疏水笼”结构为掺杂的染料分子提供了一个相对的非极性微环境,进而抑制外部水分子渗透而不会阻碍氧气分子的自由扩散,避免了染料通过电子转移途径产生的ROS对其光氧化而导致的光漂白现象。最终的实验结果表明:FSNPs相比于自由染料在水溶液中有12.3倍的荧光强度提升和增强的抗光漂白能力,并在肿瘤细胞荧光成像过程中经过连续激光扫描300秒后仍保留有90.1%的初始荧光强度。最后在2D和3D肿瘤细胞模型的PDT过程中也表现出极佳的PDT稳定性。因此,这种组合策略为合理调控FSNPs内部微环境及强化光物理性能、开发高性能FSNPs提供了新思路!
