余孝其四川大学教授。中国化学会化学生物学专业委员会委员、中国监控化学品协会技术专家库成员。是50余种国际国内重要刊物的论文评审专家,包括Angew. Chem Int. Ed.、J. Am. Chem. Soc.、J. Org. Chem.、Org. Lett.、Chem. 除了细胞屏障的基本功能外,细胞膜在许多细胞行为中发挥着非常重要的作用,如细胞迁移、细胞扩散、神经通讯、细胞运输和信号级联等。此外,细胞膜异常对于细胞状态和许多疾病而言是一个重要的标志。因此,开发高选择性、高灵敏度的检测技术以准确对细胞膜进行可视化,尤其是活体可视化监测,对于探索与解决医学早期诊断和研究生物学中的基本问题具有重要意义。
       近日,四川大学余孝其课题组与韩国高丽大学Jong Seung Kim教授课题组合作,设计了一种基于聚集诱导发光特性的荧光探针,用于生物体内细胞膜快速、灵敏的免洗成像。探针以嘌呤骨架作为核心,装配疏水的烷基链与带有正电荷的季铵盐,在水中保持良好溶解性的同时使无发光特性;探针通过疏水相互作用与静电相互作用嵌入到细胞膜中,受到磷脂双分子层的限制激活RIR过程,从而可以被激发出明亮的荧光。
探针可以对多种细胞的细胞膜进行快速的免洗成像,并且在保持高信噪比的情况下较长时间的标记细胞膜;其优良的细胞膜靶向性与生物安全性使其可以快速无损的标记神经元细胞的细胞膜。此外,探针在多细胞肿瘤球中表现出良好的细胞间渗透性与细胞膜特异性,可在免洗的情况下较快地渗透120微米的细胞球,并标记出其蜂窝状的细胞膜结构。
       除了体外染色,探针也可以对活体斑马鱼进行染色。探针可以穿透常规染料无法渗入的斑马鱼表面的粘膜层,对其表皮细胞进行染色。此外,探针也可进入斑马鱼眼组织,对其眼部结构进行部分成像。
       上研究结果以论文形式发表在国际期刊《Angewandte Chemie International Edition》上,论文题目为“An AIE-Based Probe for Rapid and Ultrasensitive Imaging of Plasma Membranes in Biosystems”。论文的第一作者为四川大学化学学院博士生石磊,通讯作者为李坤教授、Jong Seung Kim教授和余孝其教授。近期,该课题组还深入研究了探针中疏水链的长度和电荷对其细胞膜靶向性能的影响(Bio-inspired assembly in a phospholipid bilayer: effective regulation of electrostatic and hydrophobic interactions for plasma membrane specific probes, Chem. Commun., 2020, 10.1039/d0cc00679c)。四川大学为本文的第一作者单位和通讯作者单位。该项研究工作得到了国家自然科学基金的经费支持。
